REDEFINE-1 (Garvey 2025, PMID 40544433) reported ~22.7% body-weight reduction over 68 weeks — but that figure is for CagriSema, cagrilintide co-administered with semaglutide, not cagrilintide by itself. REDEFINE-5 (Yamauchi 2026, PMID 42009015) extended the Phase 3 work into Japanese and Taiwanese populations.

Read the headline carefully. Phase 2 cagrilintide monotherapy reached roughly 10–11%. Anyone comparing it against a GLP-1 alone should expect a substantially smaller standalone effect than the combination number implies. Gastrointestinal adverse events characteristic of amylin-plus-incretin combinations were reported across Phase 2/3.
